为确定江苏某猪场疑似弓形虫病发病猪群的病原,本研究取患病仔猪淋巴结、肺脏、肝脏、心和脑等组织制作常规石蜡切片,进行HE染色和免疫组化染色。将仔猪的淋巴结、肺和肝组织混合用盐酸.胃蛋白酶溶液消化处理后接种ICR鼠,经连续传代分离弓形虫,采用特异PCR方法对分离的虫株进行鉴定。结果显示在淋巴结和肺脏等组织中可见弓形虫假包囊,免疫组化染色呈强阳性;盲传第3代小鼠腹水可见弓形虫速殖子,而且通过PCR方法可以扩增得到其约301bp的Bl基因特异片段。结果表明,本研究分离鉴定出一株弓形虫虫株,命名为YZ株。
In order to confirm the pathogen of a case suspected toxoplasmosis in piglet from a pig farm in Jiangsu province, a Toxoplasma gondii was isolated by inoculation in mice and identified by specific PCR. The results showed that Toxoplasma false cysts were visible in lymph nodes and lungs; T.gondii was found out in 3rd passage of mouse ascites, and reisolated T.gondii was confirmed by PCR. The results indicated that the T.gondii was isolated and identified which was disgnated YZ strain.
