目的探讨益肾活血胶囊提取液(YSHXC)对同型半胱氨酸(HCY)诱导的兔血管平滑肌细胞(VSMCs)转化生长因子β1(TGF-β1)表达的影响。方法组织贴块法体外原代培养兔VSMCs,由HCY诱导建立细胞增殖模型并分组:正常组、HCY刺激组(HCY1mmol/L)、HCY+叶酸组(HCY1mmol/L+叶酸5mmol/L)、HCY+YSHXC大剂量组(HCY1mmol/L+YSHXC5000mg/L)、HCY+YSHXC小剂量组(HCY1mmol/L+YSHXC500mg/L)。分别采用免疫组化及RT-PCR技术检测各组TGF-β1蛋白及mRNA的表达,同时通过MTT比色法测定细胞增殖活度。结果HCY刺激后细胞数目明显增多,细胞内TGF-β1表达显著增强(P〈0.01),而YSHXC大、小剂量组细胞增殖活度减低,TGF-β1蛋白和mRNA的表达水平均下降(P〈0.01),并呈现剂量依赖性(P〈0.05),且此抑制作用优于叶酸组(P〈0.01或P〈0.05)。结论YSHXC可明显抑制HCY诱导的细胞内TGF-β1的表达,减少平滑肌细胞的增殖,这可能对延缓动脉粥样硬化进程起到一定的作用。
Objective To explore the effect of extracting solution of capsule of tonifying kidney activating blood circulation (YSHXC) on TGF-β1 expression of vascular smooth muscle cells (VSMCs) in the rabbits induced by homocysteine (HCY). Methods Primary VSMCs cultured by attachment-block in vitro were induced by HCY to establish cell multiplication model and divide into normal, HCY stimulation (HCY 1 mmol/L), HCY + folic acid (HCY 1 mmol/L + folic acid 5 mmol/L), HCY + high dosage of YSHXC (HCY 1 mmol/L + YSHXC 5 000 mg/L) ,HCY + low dosage of YSHXC (HCY 1 mmol/L + YSHXC 500 mg/L) groups. TGF-β1 protein and mRNA expressions were respectively detected by immunohistochemisty and RT-PCR. Activity of cell multiplication was determined by MTr colorimetric technique. Results After HCY stimulation, cell number obviously increased, TGF-β1 expression significantly enhanced ( P 〈 0. 01 ), but high and low dosage of YSHXC groups had the decreased activity of cell multiplication ,the descended TGFβ1 protein and mRNA expressions ( P 〈 0. 01 ), presenting dose dependent ( P 〈 0. 05 ), the better effect than folic acid group ( P 〈 0. 01 orP 〈 0. 05 ). Conclusions YSHXC could obviously inhibit TGF-β1 expressions induced by HCY,decrease VSMCs proliferation,which might be play a certain role in delaying atherosclerosis process.
