目的:应用中心点设计法,比较麻杏甘石汤主要有效成分不同配伍的药效,寻找最佳配伍,探讨各成分间相互关系。方法:以20μmol/L磷酸组胺诱发豚鼠离体气管环收缩痉挛,按照中心点设计,将麻杏甘石汤主要有效成分麻黄碱(X1)、苦杏仁苷(X2)、甘草酸(X3)组成不同配伍作用于气管环,记录其对气管环的舒张作用;运用多元非线性逐步回归法,建立回归方程,找出药效的最佳配伍,分析成分之间的相互关系。结果:得到回归方程为:Y=639.79+90.86X1+148.12X2-14.85X3+15.66X1X2-3.77X1X3-4.28X2X3+2.66X12+7.69X22+2.67 X32;盐酸麻黄碱与舒张百分率存在显著的相关性,而苦杏仁苷、甘草酸则与舒张百分率无显著的相关性;盐酸麻黄碱与苦杏仁苷对舒张百分率有非常显著地协同作用;杏仁苷与甘草酸具有显著的拮抗作用,麻黄碱与甘草酸交互作用不明显。有效成分最佳配伍为麻黄碱1.95×10-5mol/L,苦杏仁苷1.91×10-4mol/L,甘草酸1.29×10-6mol/L;成分对药效的贡献大小为:盐酸麻黄碱〉苦杏仁苷〉甘草酸。结论:中心点设计适用于麻杏甘石汤主要有效成分的配伍关系及成分相互关系的研究。
Objective: To compare the main efficacy of different combinations with effective components of Maxingganshi Decoction,to look for the best compatibility,and to explore the relationship among the components by applicating Central Composite Design. Methods: According to CCD,the ephedrine hydrochloride,amygdalin,and glycyrrhizic acid were mixed by CCD table,and applied the mixture to isolated tracheal specimens. Used Designer expert 8. 0. 1 for statistical analysis,the regression equation was built,and the compatible relationship optimization in the main active ingredient of MXSGD also was explored by multiple nonlinear stepwise regression statistical analysis. Results: The equation was established as: Y = 639. 79 + 90. 86X1+ 148. 12X2- 14. 85X3+ 15. 66X1X2-3. 77X1X3- 4. 28X2X3+ 2. 66X12+ 7. 69X22+ 2.67X32. There was highly significant correlation between Ephedrine hydrochloride and diastolic percentage,and no significant correlation between glycyrrhizic acid,Amygdalin and diastolic percentage. Ephedrine hydrochloride and amygdalin has significantly synergistic effect on diastolic percentage. Amygdalin and glycyrrhizic acid on diastolic percentage has a significant antagonism,ephedrine and glycyrrhizic acid on diastolic percentage interaction is not obvious. The effective components for the best compatibility of ephedrine 1. 95 × 10-5mol / L,amygdalin1. 91 × 10-4mol / L,glycyrrhizic acid 1. 29 × 10-6mol / L. The contribution of components on the pharmacodynamics,ephedrine hydrochloride,amygdalin,glycyrrhizic acid. Conclusion: CCD apply to research the compatibility of the main efficacy components of Maxingshigan Decoction and coorelationship ofits components.
