目的:研究中国小型猪痰瘀互结证冠心病模型血清蛋白表达谱的改变情况。方法:在建立中国小型猪痰瘀互结证冠心病模型基础上,应用双向电泳分离小型猪血清蛋白;通过图谱分析软件和基质辅助激光解析-飞行时间质谱(MALDI-TOF-MS)技术分别进行差异比较和质谱鉴定及数据比对。结果:小型猪痰瘀互结证冠心病模型组血清与正常组相比共有17个差异蛋白点,6个上调点,1个下调点,7个缺失点,3个特异表达的点。挖取这些差异蛋白点进行胶内酶解、MALDI-TOF质谱分析及MASCOT搜索引擎检索,鉴定出apolipoprotein E precursor(apoE)、complement component 4 binding protein(C4BP)等2种蛋白。结论:初步发现的差异蛋白可能与痰瘀互结证冠心病的形成、发生、发展相关,但是还需要通过其它方法做进一步验证。
Objective:To explore the changes of protein expression pattern of serum in mini-pig with syndrome of phlegm and blood stasis(痰瘀互结证)of coronary heart disease.Methods:Based on the model of mini-pig with syndrome of phlegm and blood stasis of coronary heart disease,the total proteins of serum from each group were separated by two dimensional gel electrophoresis(2-DE).We contrasted protein expressing pattern in normal and model group by Image Master 2D Elite 6.01 analysis software.Then the different proteins were identified by MALDI-TOF-MS.Results:Compared with normal group,17 proteins had been determined to change:6 proteins were up-regulated,1 proteins were down-regulated,7 proteins were lost and 3 proteins specifically expressed in mini-pig serum of the disease.With the search engine MASCOT,2 proteins including apolipoprotein E precursor(apoE),component4 binding protein(C4BP)were identified.Conclusions:These differential proteins may play an important role in the occurrence and development of syndrome of phlegm and blood stasis of coronary heart disease.The result will be further validated by other biological methods.
