目的:探讨磁感应热疗联合放疗在小鼠4T1乳腺癌细胞株皮下移植瘤模型中的治疗作用。方法:小鼠经皮下移植4T1乳腺癌细胞株,建立皮下移植瘤模型,待肿瘤直径达到10mm左右,然后将其分为以下4组进行处理:标准对照组(未做任何特殊处理,C组)、单纯热疗组(48±2℃10min,MIH组)、单纯放疗组(10Gyx2次,R组)、热放联合组(10Gyx1次+48±2℃10min,MIH+R组)。观察各治疗组和对照组的肿瘤体积的大小、肿瘤体积抑制率。每组分别在实验处理后24小时处死小鼠应用免疫组化检测肿瘤组织中Bcl-2和Caspase-3的表达情况。结果:热疗后的第1-4周MIH+R组的肿瘤体积抑制率分别为(64.38±17.97)%、(73.90±9.48)%、(77.43±10.71)%、(73.17±8.58)%;MIH组的分别为(40.66±21.46)%、(54.60±10.20)%、(51.22±5.05)%、(39.53±5.49)%;R组的肿瘤体积抑制率分别为(51.60±18.93)%、(57.53±9.76)%、(52.34±7.42)%、(35.42±4.14)%。与对照组相比,各治疗组均能抑制肿瘤的生长(P〈0.05)。其中以MIH+R组抑瘤作用最显著。与其它组相比,Bcl-2在MIH+R组的表达显著下降,而Caspase-3的表达显著上升,差异具有显著性(P〈0.05)。结论:磁感应热疗与放疗均能诱导肿瘤细胞凋亡,有效地抑制小鼠4T1乳腺癌细胞株皮下移植瘤的生长,而磁感应热疗与放疗的联合治疗的抑瘤作用最显著,而且降低了放疗剂量。
Objective: To investigate the effects of magnetic induction hyperthermia and radiotherapy on subcutaneous tumor of 4T1 breast carcinoma in rats.Methods: After the induction of 4T1 breast carcinoma cells,the mices were randomly assigned to four groups: the control group(group C),magnetic induction hyperthermia group 48±2℃ 10min,group MIH),radiotherapy group(10Gy x2 fraction,group R),combination group(10Gy x1 fraction +48±2℃ 10min,group MIH+R).The volume of the tumors were observed.24 hours after the treatment,3 mices of each group were killed and the tumor tissue were excised,the expression of the gene Bcl-2,Caspase-3 were determined with ABC immunohistochemical staining.Results: From the first week to the fourth week,the inhibitive rates of tumor volume of group MIH+R was(64.38±17.97)%,(73.90±9.48)%,(77.43±10.71)%,(73.17±8.58)%,those of group MIH was(40.66±21.46)%,(54.60±10.20)%,(51.22±5.05)%,(39.53±5.49)% and those of group R was(51.60±18.93)%,(57.53±9.76)%,(52.34±7.42)%,(35.42±4.14)% respectively.Both magnetic induction hyperthermia and radiotherapy could inhibit the growth of tumor,but the therapeutic effect was more significant in group MIH+R(P0.05);The expression of Bcl-2 were significantly decreased and Caspase-3 were significantly increased in the combination groups(P0.05).Conclusion: We found that both magnetic induction hyperthermia and radiotherapy could inhibit the growth of tumor,but the therapeutic effect was more significant in group MIH+R(P0.05).
