目的:研究海人酸致痫大鼠模型中P-糖蛋白(P—gP)表达水平的变化及雷公藤内酯干预对其的影响。方法:84只SD大鼠随机分成3组,其中对照组28只,海人酸致痫组28只。雷公藤内酯干预组28只。应用Western—Blot法检测各组动物海人酸致痫后6、24、48、72h海马区P-gp的表达,RT—PCR法检测各组动物各时间点P—gPmRNA的表达。结果:雷公藤内酯干预组海马区各时间点P—gp及P—gpmRNA的表达均较海人酸致痫组显著下调(P〈0.05),且雷公藤内酯干预后48h下调P—gp作用明显。结论:雷公藤内酯对海人酸致痫大鼠耐药有逆转作用,其机制与下调致痫大鼠海马区P-gp的表达有关。
Objective:To study the changes on the expression level of P-glycoprotein (P-gp) in the model of kainite-induced rat and the effect of triptolide intervention. Methods : 84 SD rats were divided into 3 groups, per group for 28 rats, named as control group, kainite-induced group and triptolide treatment group. Western-Blot was used to detect the expression of P-gp in the hippocampal region after injection kainite 6 h,24 h,48 h,72 h. RT-PCR method was used to observe rats mRNA level of P-gp each group at the same time. Results : The expressions of P-gp in the hippocampal region in triptolide treatment group were decreased significantly than those of kainite-induced group ( P〈0.05 ) and after injection triptolide 48 h P-gp was inhibited obviously. Conclusion : Triptolide can changeover the drug resistance of kainite- induced rats. Its mechanism is connected with inhibiting the expression level of P-gp in the hippocampal region in epilepsy rats.
