本文以葡萄糖氧化酶(GOx)为研究对象,采用壳聚糖(Chitosan)-单壁碳纳米管(SWCNTs)复合膜固定GOx构建一种膜体系,研究了GOx在膜上的直接电子转移和对葡萄糖的电催化。通过循环伏安法,在pH=7.0的磷酸盐缓冲溶液中对膜电极进行表征,得到一对形态良好的可逆氧化还原峰,探讨了在不同pH值的磷酸盐缓冲溶液及不同表征底液中的电子转移情况。同时研究了该体系对葡萄糖的电催化。结果表明该体系对葡萄糖的氧化具有良好的电催化作用,其线性范围为4.0×10-4~5.6×10-3mol/L,检测限为2.4×10-5 mol/L。该体系具有良好的稳定性和重现性。
A new biosensor was successfully fabricated based on the composite film of chitosan-single-walled carbon nanotubes(Chit-SWCNTs) wrapped glucose oxidase(GOx).The system aimed at studying the direct electron transfer of GOx in the composite membrane of Chit-SWCNTs.The SWCNTs were dispersed into chitosan solution firstly,and then the mixture was fixed on the surface of gold electrode(GE) at a certain volume.After the mixture on the surface of GE evaporated,the GE was immersed in the solution of GOx for 10~12 hours.A pair of well reversible redox peaks were obtained in phosphate buffer solution(pH=7.0) by characterizing the membrane electrode.Meanwhile,the direct electron transfer of GOx under different conditions of the phosphate buffer solution with different pH and different characterization of a base solution was also investigated.The Chit/SWCNTs/GOx/GE also showed good stability and reproducibility.
