为了提高物质的制备量和分离度,采用半制备液相色谱法对油茶果皮多酚的制备条件进行了优化实验。结果表明,在用50%乙醇提取油茶果皮多酚的基础上,以半制备液相色谱法分离纯化油茶果皮多酚的优化条件如下:流动相A(水∶乙酸=100∶1)和B(乙腈),以2.5 mL/min的流速进行梯度洗脱,进样量为0.4 mL。实验结果还表明:按此方法制备所得单一多酚组分含量可达21.22 mg/g。这一实验结果证明了此法有效可靠。
In order to improve preparation quantity and separation degree, the preparation condition of polyphenol in Camellia pericarp was optimized by semi-preparation HPLC. The results showed that after extracting polyphenol with 50% of alcohol, the optimization condition of isolating and purifying polyphenol by semi-preparation HPLC was as follows: gradient elution was carried on by mobile phase A (volume ratio of water to acetic acid was 100 : 1) and B (acetonitrile) at 2.5 mL/min flow rate, the injection volume was 0.4 mL. The content of single polyphenol was 21.22 mg/ g through the preparation method, and it was effective and reliable.
