为分析2009~2010年中国华东地区猪圆环病毒2型(PCV2)的基因型分布情况,应用可以区分PCV2a和PCV2b的鉴别PCR,对浙江、江苏、安徽、山东、上海、江西等6省市的210份血液和组织样品进行PCV2检测,并选取10份阳性病料进行全长基因组扩增,应用DNAStar软件对获得的10株PCV全长基因和GenBank中的参考毒株进行序列分析和系统进化树分析。结果表明210份病料中107份为PCV2阳性,36份为PCV2a基因型,97份为PCV2b基因型,检出率分别为17.1%和46.2%,两个基因型的共感染率为12.4%。10株分离株的序列分析结果表明,华东地区PCV2流行毒株之间的核苷酸同源性为96.3%~99.9%,与GenBank其他PCV2参考毒株的同源性为95.0%~99.9%。系统进化树分析表明中国华东地区PCV2存在3种基因型:PCV2a、PCV2b和PCV2c。
This study aimed to analyse the genotype distribution of porcine circovirus type 2(PCV2) in eastern China from 2009 to 2010.A total of 210 porcine sera and tissue samples obtained from Jiangsu,Anhui,Shandong,Zhejiang,Jiangxi provinces and Shanghai of China during 2009-2010 were detected using distinction PCR of PCV2a and PCV2b.In order to fully understand the genetic variation of PCV2 in recent years,the complete genome of 10 PCV2-positive templates were amplified by PCR.Their sequences were compared with the other reference strains sequences of PCV2 in GenBank to analyse the sequence and construct phylogenetic tree.The results showed that 107 of 210 samples were PCV2-positive,36 samples belonging to PCV2a genotype and 97 samples belonging to PCV2b genotype.The PCV2a and PCV2b infection rates were 17.1% and 46.2% respectively,and the co-infection rate was 12.4%.The sequence analysis result revealed that 10 isolates shared 96.3%-99.9% nucleotide sequence similarity and 95.0%-99.9% with other reference strains of PCV2 in GenBank.The phylogenetic analysis result indicated that all these isolates genotypes could be divided into three groups as PCV2a,PCV2b and PCV2c.
