目的:研究温阳解郁汤对慢性应激抑郁大鼠HPA轴的影响,初步探讨其抗抑郁作用的机制。方法:雄性Wistar大鼠,随机分为6组,正常对照组和模型对照组灌胃0.9%氯化钠溶液,温阳解郁汤高、中、低剂量组灌胃温阳解郁汤浓缩煎剂(8、6、4g/kg)、阳性药对照组灌胃盐酸氟西汀(3.33mg/kg),除正常对照组外其余各组在灌胃同时接受慢性不可预知温和刺激(CUMS)刺激并孤养28d。采用强迫游泳实验(FST)、悬尾实验(TST)、旷场实验(OFT)对各组大鼠进行行为学评价;q PCR检测大鼠下丘脑CRH m RNA和海马GR m RNA的表达;放射免疫法检测大鼠血中ACTH、CORT的含量;HE染色大鼠海马观察组织形态变化。结果:温阳解郁汤能够减少大鼠FST和TST不动时间,增加OFT穿格次数(P〈0.05);能够降低由慢性应激引起的CRH、GR、ACTH、CORT水平升高(P〈0.05),并能减少海马组织损伤。结论:温阳解郁汤能够改善CUMS大鼠的应激抑郁行为和慢性应激时高糖皮质激素对大脑的损害,其作用机制可能与调节HPA轴有关。

英文摘要:

Objective: To study the influence of Wenyang Jieyu Decoction on HPA axis of depression rats, and investigate the mechanism of antidepressant of Wenyang Jieyu Decoction. Methods: Male wistar rats were randomly divided into 6 groups as control group, model group, high, middle and low dosage of Wenyang Jieyu Decoction groups and l ioxetine group. Rats in all the groups except control group were received gavage administration and chronic unpredictable mild stress(CUMS) combined with isolation stimulus simultaneously for 28 days. The behavioristics of rats in each group were evaluated by using forced swimming test(FST), tail suspension test(TST) and open field test(OFT). The expression of CRH m RNA in hypothalamus and GR m RNA in hippocampus of rats were determined by using q RCP method, and the contents of ACTH and CORT in serum of rats were determined by using radioimmunoassay. The histomorphology changes were observed by using HE staining. Results: Wenyang Jieyu Decoction could reduce the immobilization time of rats in FST and TST, and increase the cross number of rats in OFT(P〈0.05). It also could decrease the levels of CRH, ACTH and CORT of rats caused by CUMS(P〈0.05), and reduce the damage of hippocampal tissue. Conclusion: Wenyang Jieyu Decoction can improve the depression behaviors of CUMS rats and brain damages caused by high sugar cortical hormone, and the mechanism may relate with the regulation effects on HPA axis.
