目的:探讨固本化痰通脉方对痰阻血脉大鼠血脂代谢及核转录因子-κB(NF-κB)表达的影响.部分揭示痰邪的致病机理。方法:6~7周龄正常雄性Wistar大鼠40只随机分为正常组、模型组、中药固本化痰通脉方组、西药对照组4组,透射电镜下观察主动脉内皮的超微结构,采用免疫组织化学法检测核转录因子-κB(NF-κB)的表达。结果:与正常组相比,模型组血脂水平、NF-κB的表达有明显差异(P〈0.01);各治疗组与模型组相比有差异(P〈0.05)或显著差异(P〈0.01);电镜观察固本化痰通脉方组主动脉内皮更接近正常。结论:固本化痰通脉方能够降低痰阻血脉大白鼠血清胆固醇、甘油三酯的含量,改善动脉硬化和延缓动脉硬化的发展。其机制可能与抑制NF-κB活性.降低细胞黏附分子的表达有关。
Objective: To explore the effects of Guben Huatan Tongmai Prescription(GBHTYMP) on metabolism of blood lipid and nuclear factor-κB in rats with blood vessel blocked by phlegm, and partially disclose the pathogenesis of phlegm - pathogen. Methods: Forty normal male Wistar rats,6 - 7month in age, were randomly divided into four groups: normal group, model group, traditional Chinese medicine group and control group. Establishing the model of phlegm symptoms based on the method of feeding with high lipid diet. Contrasted the pathological changes in blood lipid with each other,observed the ultrastructural patho- logical states of vascular endothelism with transmission electron microcope. Macrophage and nuclear factor-κB by immunohisto- chemistry analysis. Results : Contrasted to the normal group, model group showed significant differences on the level of blood lip- id, and nuclear factor-κB ;The two treatment groups had difference( P 〈0.05) or obvious difference with model group( P 〈 0.01 ). The states in vascular endothelism of traditional Chinese medicine group were nearly normal. Conclusions: GBHTTMP can effectively decrease the level of TCh, TG, LDL - Ch and increase the expression of HDL - Ch. The prescription also can reduce atherosclerotic progression to some extent. The mechanism of anti - atherosclogentic by GBHTTMP may be related to inhibition of nuclear factor-κB and cell adhesion molecules.
