目的探讨蛋白激酶PI3K/Akt信号途径在肝组织再生中的作用.方法采用切除小鼠肝组织2/3的部分切除法,制成小鼠肝再生模型;小鼠体内肝组织P13K的活性用腹腔注射Wortmannin来抑制.分别采用western blot和RT-RCR方法检测Akt磷酸化水平和增殖细胞核抗原PCNA mRNA表达.结果小鼠肝组织部分切除48h后,PCNA基因表达显著升高,表明肝组织再生发生.小鼠肝组织部分切除24h后,Akt磷酸化水平明显升高.PI3K特异性抑制剂wortmannin显著抑制Akt磷酸化水平和PCNA基因表达.结论PI3K/Akt信号途径在小鼠肝组织再生过程中起重要作用.
Objective To study the role of PI3K/Akt pathway in liver regeneration in mice. Methods The mouse model of liver regeneration was made by two-third partial hepatectomy (PH). To inhibit PI3K activity in mice, wortmannin was given intraperitoneally. The phosphorylation of Akt was measured by Western blotting; The mRNA levels of proliferation cell nublear antigen PCNA were detected by RT-PCR. Results The expression of PCNA markedly increased after 48 h of PH, indicating that the liver regeneration oecured. The pho- sphorylation of Akt increased after 24 h of PH. Wortmannin, a specific inhibitor of PI3K significantly inhibited the phosphorylation of Akt as well as the expression of PCNA. Conclusion PI3K/Akt pathway plays an important role in liver regeneration in mice.
