建立了化学-海因酶法制备光学纯D-茶氨酸的新方法。采用氰酸盐和L-谷氨酸关环制备D,L-羧乙基海因,与乙胺进行侧链衍生后得到酶转化底物N-乙基海因丙酰胺;进而采用D-海因酶催化水解得到N-氨甲酰-D-茶氨酸,经亚硝酸水解最终得到标题化合物。总收率达55%。对海因酶催化和亚硝酸水解过程条件进行了优化。
A novel preparation method of D-theanine by chem- hydantoinase was studied. First,D ,L-5-carboxyethyl hydantoin was produced using cyanate and L-glutamatic acid as material with ring closing in the alkaline condition,and N-ethyl-3-(2, 5-dioxoimidazolidin-4-yl) propanamide was got sequentially by side chain reaction of D, L-5-carboxyethyl hydantoin. Then, the N-ethyl-3-(2,5-dioxoimidazolidin-4-yl) propanamide was hydrolyzed to N-carbamoyl-D-theanine by D-hydanionase,and D-theanine was got by hydrolysis of N-carbamoyl-D-theanine with nitrous acid. The total yield of D-theanine was 55%. The conditions of enzymatic reaction and decarbamyl reaction was also optimized.
