目的研究蒿甲醚对约氏疟原虫在大劣按蚊体内发育的影响及其机制。方法本课题利用已经建立的斯氏按蚊-约氏疟原虫模型,通过吸饲蒿甲醚糖水的方法,观察蒿甲醚是否影响约氏疟原虫在斯氏按蚊体内的发育。通过RT-PCR方法,分析NOS、TEP1和PPO三个斯氏按蚊重要的免疫反应相关基因的表达变化,以了解青蒿素类药物蒿甲醚影响疟原虫在按蚊体内的发育机制。结果蒿甲醚可以促进疟原虫在按蚊体内的发育,使卵囊数量增长3-5倍。对其机制的研究中发现,斯氏按蚊吸饲蒿甲醚后,使NOS、TEP1和PPO三个免疫相关基因在转录水平下调,提示蒿甲醚可能通过抑制NO产生,丝氨酸级联反应及溶解反应,降低按蚊免疫反应,促进疟原虫在按蚊体内发育。结论蒿甲醚可能通过抑制斯氏按蚊免疫相关基因的表达,促进约氏疟原虫在斯氏按蚊体内发育。
Objective To explore the effect of artemether on the development of Plasmodium yoelii in Anopheles stephensi and its mechanism.Methods Basing on Plasmodium yoelii / Anopheles stephensi model,mosquitoes were fed on artemether,which the effect of artemether on the development of Plasmodia was observed.To learn the mechanism of artemether effected the development of Plasmodia,which the transcript variation of NOS,TEP1and PPO was analyzed by using RT-PCR.NOS,TEP1and PPO play important roles in immune response against plasmodia infection.Results Artemether enhanced the development of Plasmodium yoelii in Anopheles stephensi,and made the oocysts increase 3-to 5-folds.RT-PCR results showed that artemether inhibited the expression of NOS,TEP1 and PPO.Our research suggested that artemether may weaken Anopheles stephensi immune response against Plasmodium by inhibiting NO synthesis and melanization or lysis,which resulted in better development of Plasmodium in Anopheles stephensi.Conclusion Our research suggested that artemether may inhibit the expression of immune genes of mosquites,which results in better development of Plasmodium yoelii in Anopheles stephensi.
