目的观察蛴螬提取物对兔视网膜静脉阻塞(retinal vein occlusion,RVO)模型ET-1表达的干预作用。方法取健康兔36只,每组9只,分为空白组、模型组、血栓通组、蛴螬组4组O采用波长532nm的Nd:YAG激光光凝法建立RVO模型,连续灌胃给药,实验后7、14、21d分别处死兔取材,石蜡包埋切片,用免疫组化法检测视网膜组织中ET—1表达活性,计算机图像分析系统并进行统计分析。结果蛴螬组7、14、21d与模型组比较,ET-1表达明显下降,差异有统计学意义(P〈0.05);蛴螬组与血栓通组比较,差异无统计学意义(P〉0.05);蛴螬组与空白组比较,7、14d的ET-1表达活性明显高于空白组,差异有统计学意义(P〈0.05),21d时的ET-1表达活性与空白组差异无统计学意义(P〉0.05)。结论蛴螬能够明显降低兔RVO模型ET-1表达。抗血栓形成。改善RVO后视网膜局部微循环,减轻缺血缺氧对血管内皮细胞的损害。
Objective To observe the effects of grub extract on the expression of retinal ET-1 of rabbits' retina vein obstruction model. Method 36 healthy rabbits were randomly divided into 4 groups as follows: blank group, modeling group, xueshuantong group and grub group, 9 rabbits in each group. RVO models were made with Nd: YAG laser photocoagulation whose wavelength is 532 nm, administered intragastrically continuously. The rabbits were executed respectively in the 7th, 14th and 21st days after the treatment, taking the rabbits retinal tissues and embedding in paraffin. The ET-1 expression levels of the retinal tissues were tested with immunohistochemistry, and then results were analyzed in computer image the modeling group, the ET-1 expression levels in grub analysis system. Results Compared with group decreased markedly in 7 d, 14 d and 21d three time points (all P〈0.05). But there were no significant differences between xueshuantong group and grub group (P〉0.05). The ET-1 expression levels in grub group were significantly higher than those in blank group in 7d, ldd two time points (all P〈0.05), but there were no significant differences in 21d (P〉0.05). Conclusion Grub can significantly decrease the expression of ET-1 in rabbits' RVO model, has anti-thrombosis effects, can improve the RVO local microcirculation, and alleviate the injury resulted from hypoxia-ischemia on vascular endothelial cells.
