目的:探讨天麻素对星形胶质细胞增殖及胶质细胞源性神经营养因子(GDNF)释放水平的影响.方法原代培养海马星形胶质细胞,将其随机分为天麻素组和对照组.天麻素组细胞分别直接给予终浓度为10,15,20,25,50μg/ml 的天麻素,处理48 h 或72 h.采用细胞计数试剂盒(CCK-8)检测星形胶质细胞活性,5-溴脱氧尿嘧啶核苷(BrdU)检测细胞增殖情况,酶联免疫吸附测定(ELISA)检测 GDNF 释放水平.结果48 h 后各组的星形胶质细胞活力、BrdU 阳性细胞数量以及 GDNF 的释放水平差异均无统计学意义(P >0.05);72 h 后各组的星形胶质细胞活力、BrdU 阳性细胞数量以及GDNF 的释放水平差异均有统计学意义(P <0.05),进一步两两比较,20μg/ml 和25μg/ml 天麻素组的上述3个指标均高于其他剂量组和对照组(P <0.05).结论一定剂量的天麻素能够促进星形胶质细胞的活性和增殖,并且上调 GDNF 的释放水平.
Objective To investigate the effect of gastrodin on the proliferation of astrocytes,as well as the release of GDNF.Methods Astrocytes derived from hippocampal of fetal rats were divided in-to gastrodin group and control group.In gastrodin group,the astrocytes were treated with different con-centrations of gastrodin (10μg/ml,1 5 μg/ml,20μg/ml,25 μg/ml and 50μg/ml)about 48 h or 72 h.The cells viability,the BrdU labeling and detection and the level of GDNF were measured by the kit of CCK-8, immunocytochemical method and Elisa kit,respectively.Results The cell viability,the number of BrdU+cells and the level of GDNF in 20 μg/ml and 25 μg/ml gastrodin treated groups were significantly in-creased compared to the control and other groups after 72 h incubation (P 〈 0.05).However,there were no significant differences between each group after 48 h incubation.Conclusions The proliferation and re-lease of astrocytes and the level of GDNF can be elevated by gastrodin.
