目的: 观察适时适量补充硒(Se) 和维生素E(VE) 对心肌纤维化和心肌重构的影响, 探讨克山病病区的低Se和低VE膳食是否在心肌损伤后的修复过程中持续发挥作用。方法:30只Wistar大鼠随机分为: 正常对照组(Control, 混合饲料喂养) 、病区粮饲料组(EG, 病区粮喂养) 、病区粮饲料加Se组(EG + Se) 、病区粮饲料加VE组(EG+ VE) 和病区粮饲料加Se加VE组(EG+ Se + VE) 。喂养21 d后, 取大鼠心脏, 比色法测定大鼠血清中谷胱甘肽过氧化物酶(GSH- Px) 活力; 硫代巴比妥法测定血清丙二醛(MDA) 含量; 马松三色染色测定大鼠细胞外基质沉积情况; 免疫组织化学法和Western blotting法测大鼠心肌组织中转化生长因子β(TGF-β) 和结缔组织生长因子(CTGF) 表达。结果: 与正常对照组比较,EG组心肌纤维化大鼠血清中GSH- Px活力下降(P <0. 01) ,MDA含量升高(P <0. 05) , 心肌组织胶原蛋白大量沉积, 并伴有TGF-β和CTGF表达上调(P <0. 01) 。与EG组比较,EG+ Se组和EG+ Se + VE组病区粮心肌纤维化大鼠血清中GSH- Px活力均提高(P <0. 01和P<0. 05) ,MDA含量降低(P <0. 05) ; 大鼠心肌细胞外基质沉积缓解; 克山病大鼠心肌TGF-β和CTGF表达下调(P <0. 01) 。结论: 低Se和低VE在心肌损伤后的修复过程中持续发挥作用, 适时适量补充Se和VE有利于减轻和逆转过度的心肌纤维化和心肌重构。
Objective To observe the effects of adequate seleniu m(Se)and vita min E(VE)on myocardial fibrosisand re modelling and explore the effects of Se and VE in the repair process after myocardial injury in Keshan diseasearea . Methods Thirty Wistar rats were randomly divided into control group(fed on mixed grain) ,Keshan diseasearea diet group(EG,fed on Keshan disease area diet continuously) ,Keshan disease area diet plus Se group(EG+Se) ,Keshan disease area diet plus VE group(EG+ VE)and Keshan disease area diet plus Se and VE group(EG+Se + VE). All rats were sacrificed after 21 d . The activitiy of glutathione peroxidase(GSH- Px)was detected bycolori metry;the expression of malondialdehyde(MDA)in the seru m of rats was detected by TBA. The depositionof extracelluar matrix in different groups was detected by mass on trichrome staining . The expressions of TGF-βand CTGF in myocardial cells of rats were detected by i mmunohistoche mical staining and Western blotting method .Results In the myocardial fibrosis rats fed on kashan disease area diet,the seru m GSH- Px activity was decreased,the level of seru m MDA was increased,myocardial fabric accu mulation and the expression levels of TGF-βandCTGF were up-regulated . Compared with EG group,the seru m GSH- Px activities wre increased(P 〈0. 01 and P 〈0. 05) ,the MDA contents were decreased(P 〈0. 05) ,the cardiac extracellular matrix deposition and theexpressions of TGF-βand CTGF in rat myocardiu m were decreased(P 〈0. 01)in EG+ Se group and EG+ Se + VEgroup . Conclusion Low Se and VE play a continuing role in the repair process after myocardial injury . Theadequate supple mentation of Se and VE is able to reverse the myocardial fibrosis and re modelling .
