目的 检测酸敏感离子通道(ASICs)在大鼠佐剂性关节炎关节软骨中的表达情况。方法大鼠右侧后足跖皮内注射弗氏完全佐剂(CFA)诱导佐剂性关节炎(AA)大鼠模型,用半定量反转录-聚合酶链反应(RT—PCR)和Westernblot方法检测ASICs在AA大鼠关节软骨的表达。结果经半定量RT—PCR分析,ASIC1a、ASIC2a和ASIC3在AA大鼠关节软骨中的表达量明显高于正常组.差异有统计学意义(P〈0.01);Westernblot进一步检测显示蛋白表达的变化与mRNA变化一致。结论AA大鼠关节软骨中ASICs的表达增多,可能与关节软骨破坏有关。
Objective To study the expression and significance of acid-sensing ion channels (ASICs) in rat articular cartilage with adjuvant arthritis. Methods Complete Freund's adjuvant(CFA) was prepared by suspending beat-killed Bacillus Calmette Guerin (BCG) in liquid paraffin at 10 mg/ml. CFA-induced arthritis was developed by injection of 100μl CFA emulsion intradermally into the right hind paw. The morphological changes of articular tissues was observed by light microscope; RT-PCR and immunoblotting analyses were used to detect ASICs in rat articular cartilage with adjuvant arthritis. Results RT-PCR and western blot showed that ASICla, ASIC2a and ASIC3 were present in the articular cartilage of normal and model group, the ASICs mRNA levels in the model group were higher than in the normal group detected by semiquantitative analysis (P〈0.01), ASICs protein levels in model group were higher than those in the normal group (P〈0.01) when examined by immunoblotting. Conclusion The results show that the expression of ASICs in AA articular cartilage is enhanced and it may be related with articular cartilage breakdown.
