[目的]分析甘草紫杉烯5α-羟化酶(GuT5H)cDNA序列及其编码的蛋白质的结构特征.[方法]以甘草GuT5H全长cDNA为研究对象,运用生物信息学软件和网络工具预测其编码的蛋白质序列,并分析该蛋白质的结构、功能及分子进化特征.[结果]甘草GuT5H cDNA全长1692 bp,含有1428 bp的开放阅读框,编码475个氢基酸残基的蛋白质;该蛋白质与细胞色素P450家族紫杉烯5αt-羟化酶(T5H)成员具有高度的序列一致性,分子进化上与同科植物大豆、苜蓿、鹰嘴豆的T5H关系最为密切.[结论]甘草GuT5H cDNA编码紫杉烯5α-羟化酶,属于细胞色素P450家族成员.
[Objective] To analyze the structure characteristics of a taxadiene 5α-hydroxylase cDNA (GuTGH) and the deduced amino acid sequence in Glycyrrhiza uralensis. [Methods] The completed cDNA sequence and the structure, function, phylogeny of the deduced amino acid sequence of GuT5H were predicted by bioinformatics analysis. [Results] The cDNA sequence of GuT5H was 1692 bp with an open reading frame of 1428 bp, encoding a protein of 475 amino acid residues. The GuT5H protein has a high sequence identity with other taxadiene 5α-hydroxylases (T5H) which belong to the cytochrome P450 family. Phylogenetically, GuT5H was closely related with T5Hs in Glycine max, Medicago truncatula, Cicer arietinum and other Leguminosae plants. [Conclusion] GuT5H cDNA encode a protein of taxadiene 5α-hydroxylase, which belong to the cytochrome P450 family.
