本研究采用RAPD技术对10个不同的白灵菇株系进行遗传多样性分析。经过筛选得到的4种随机引物共扩增出52条DNA片段,其中多态性条带44条,多态性位点频率为84.6%,说明通过RAPD扩增获得的白灵菇10个株系的遗传指纹图谱多样性丰富。利用Popgen32生物学软件计算各株系间遗传距离并构建系统进化树,在分子水平上确定了10个白灵菇株系间的亲缘关系,可为进行更科学、准确分类提供参考依据。
Diversity of genomic DNA of ten cultivated strains of Pleurotus nebrodensis was analyzed by RAPD (random amplified polymorphic DNA). 52 DNA fragments were amplified from 4 selected primers, among which 44 were polymorphic bands with 84.6% of polymorphic site frequency.
