从四川感病的辣椒上分离得到了辣椒脉斑驳病毒(Chilliveinalmottlevirus,ChiVMV)的一个分离物Pp3.利用RT—PCR(reversetranscriptionpolymerasechainreaction)的方法克隆这个分离物的外壳蛋白(coatprotein,CP)基因,测定了它的核苷酸序列并进行序列分析.结果表明Pp3的CP基因含有861个碱基,与迄今已报道的其它辣椒脉斑驳病毒分离物CP基因的序列一致性为80.0%-,88.ooA.基于辣椒脉斑驳病毒CP基因核苷酸序列的分子进化树显示,Pp3与其它分离物的差异较为显著.利用模式植物烟草对Pp3的致病性特征进行了初步的研究,结果显示Pp3具有较强的致病性,能导致烟草产生系统性坏死症状.
One isolate (Pp3) was isolated from infected pepper in Sichuan province. The isolate was iden- tified as Chilli veinal mattle virus (ChiVMV). The CP (coat protein) gene of the isolate was cloned by RT-PCR (reverse transcription polymerase chain reaction) and was sequenced and compared with the CP gene sequences of other ChiVMVs reported in GenBank. The results showed that the sequence of the CP gene was 861 nucleotides in length. Sequence alignment showed that this isolate shared 80.0%~88.0~ identity in the CP nucleotide sequence level with those ChiVMV isolates obtained from other parts of the world. Based on the phylogenetic analysis of the CP gene of ChiVMVs, it showed that the variation of isolate Pp3 was evident. Using model plant Nicotiana tabacum the pathogenicity of this virus was studied preliminarily. The results revealed that Pp3 had higher pathagenicity and could induce systemic necrosis in N. tabacum.
