在对酶-表面活性剂超活性和反胶束酶反应器的研究中,表面活性剂对酶催化动力学的影响是定量研究酶活性的关键之一.以α-糜蛋白酶作为模型酶,研究了阳离子表面活性剂GN10存在下对醋酸二萘酯的催化水解.由于表面活性剂与酶的弱相互作用,使酶催化动力学发生改变.对酶水解的初始速率进行了讨论,为酶-表面活性剂体系的酶活性研究提供了定量测量的实验方法和理论依据.
In the study on catalytic superactivity of enzyme-surfactant and enzymatic reactor with reverse micelle,the effect of surfactants on the catalytic kinetics is important for assessing quantitatively the enzyme activity.The catalytic hydrolysis of 2-Naphthyl Acetate(2-NA)byα-chymotrypsin(α-CT)was studied by spectrophotometry in the presence of cationic GN10.The initial rate and linear period of hydrolysis reaction of 2-NA varied possibly due to the change of the subtle conformation ofα-CT,depending on the weak interaction between surfactant andα-CT.The quantitative methodology and theoretical knowledge aboutα-CT activity in aqueous surfactant environment were evidenced.
