目的:研究淫羊藿苷(Icariin,ICA)对快速老化小鼠SAMP8学习记忆功能的影响及与血清雌激素水平相关性。方法:按体重随机挑选10只6月龄抗快速老化小鼠SAMR1和40只同龄快速老化小鼠SAMP8,并将SAMP8随机分为4组(模型组、己烯雌酚组和ICA 75 mg.kg-1、150 mg.kg-1组),灌胃给药,3个月后,用Morris水迷宫法检测SAMR1和SAMP8的学习记忆能力;放射免疫法检测血清雌二醇(Estradiol,E2)水平;处死称量子宫湿重,计算子宫系数。结果:ICA可以改善SAMP8定位航行和空间搜索学习记忆障碍,提高血清E2水平,但对子宫系数影响不明显;Pearson相关系数分析发现ICA对SAMP8 Morris水迷宫潜伏期的影响与血清E2水平呈负相关的关系(R=-0.64,P=0.003)。结论:ICA可以改善SAMP8的学习记忆能力,其作用机理可能与升高血清雌激素水平有关。
Objective:To probe the correlation between the effect of icariin on learning and memory and the level of estadiol(E2 ) in serum in senescence accelerated mouse prone/8 (SAMP8). Methods: 10 six-mon-old senescence accelerated mouse resistance 1 (SAMR1) and 40 the same age SAMP8 were selected. SAMP8 were divided randomly into 4 groups (model, 75 and 150 mg·kg^-1 icariin ig, and 0.2 mg·kg^-1 diethylstilbestrol ig). Then, the effects of icariin administration by oral gavage for 3 months on learning and memory deficits in SAMP8 and SAMR1 were investigated by the Morris water maze test; the level of E2 in serum in SAMP8 and SAMR1 was determined by radioimmunoassay kit. The mice were sacrificed after ethology test, and the uterus coefficient was calculated. Results: The administration of icariin orally could enhance the learning and memory ability of SAMP8 in Morris water maze test; increase the level of E2 in serum in SAMP8; create non-significant difference in the uterus coefficient of SAMP8. There was a significant negative correlation between the latency in Morris water maze test and the level correlation coefficient. Conclusions: Icariin could improve the cognitive of E2 in serum impairments of in SAMP8 analyzed by Pearson SAMP8, and its mechanism may be related to the increase in the level of estrogen in serum in SAMP8.
