目的研究Celecoxib(塞来考昔)对人黑素瘤A375细胞增殖抑制、凋亡诱导和周期阻滞作用及时A375细胞表达Cox-2和分泌型MMP-1蛋白的影响。方法MTT法测定Celecoxib对A375细胞的增殖抑制作用,流式细胞术检测药物诱导细胞凋亡及周期阻滞作用;细胞爬片SABC免疫细胞化学检测Celecoxib对A375细胞Cox-2蛋白表达的影响,ELISA法分析Celecoxib对A375细胞分泌型MMP-1蛋白表达的影响。结果80μmol/L的Celecoxib明显抑制人黑素瘤A375细胞增殖,并诱导凋亡(凋亡率35,91%)和G2/M期阻滞,减少A375细胞表达Cox-2蛋白和分泌型MMP-1蛋白。结论人黑素瘤A375细胞表达Cox-2蛋白和MMP-1蛋白,Celecoxib是-种有潜力的抑制黑素瘤增殖的药物。
Objective To investigate the effects of Celecoxib on cell proliferation, apoptosis, cycle arrest and Cox-2/MMP-1 protein expression of human melanoma A375 cell. Methods MTT assay was used to determine the anti-proliferative effects of Celecoxib on human melanoma A375 cell. Flow cytometry was performed to investigate the influence of Celecoxib on cell cycle and cell apoptosis. SABC immunocytochemistry was employed for the expressions of Cox-2 protein expressions. ELISA was used to detect secreting MMP-1 protein expression. Results 80μmol/L Celecoxib markedly inhibited the proliferation and induced apoptosis (apoptosis ratio =35.91% ) and GJM cycle arrest on human melanoma A375 cell. Cox-2 protein and secreting MMP-1 protein were expressed in human melanoma A375 cell. Celecoxib could significantly inhibit the expressions of Cox-2 protein and secreting MMP-1 protein. Conclusion Human melanoma A375 cells express Cox-2 protein and MMP-1 protein. Celecoxib may be a potential chemotherapeutics for the proliferation of melanoma.
