黑米种皮中花色苷组成及含量的HPLC分析

花色苷是黑米种皮中重要的活性成分,具有多种生物活性,准确分析其组成及含量对黑米的加工利用研究有重要意义。采用酸性甲醇溶液浸提黑米种皮得到粗提液,用高效液相色谱法(HPLC)通过与标准品对照,分析黑米皮中主要花色苷的组成种类及含量,旨在建立一种简便、准确的黑米种皮花色苷分析方法。研究结果表明,黑米种皮中主要含有3种花色苷组份,分别是矢车菊-3-葡萄糖苷(23.30 mg/g),矢车菊-3-芸香糖苷(0.97 mg/g),芍药定-3-葡萄糖苷(2.24 mg/g)。该方法样品前处理简单,是一种简便、准确的花色苷组成及含量的测定方法。

英文摘要:

Anthocyanins,which have shown many health-promoting activities,are most important components in black rice pericarp.The purpose of this study is to establish a simple method to determine the anthocyanins composition and contents in black rice pericarp.Anthocyanins crude extract solution was obtained by soaking black rice pericarp in acidified methanol.The HPLC gradient elution method was used to analyze anthocyanins composition and contents.The identification and peak assignment of anthocyanins were primarily based on comparison of their retention times with those of standards and literature.The content of each anthocyanin was calculated by HPLC peak area compared with external standard calibration curve.Results:The identified anthocyanins from black rice pericarp are cyanidin-3-glucoside(23.30 mg/g),cyanidin-3-rutinoside(0.97 mg/g),and petunidin-3-glucoside(2.24 mg/g).The method established in this experiment,requiring a simple sample pre-treatment,is a quick and accurate method to determine the composition and contents of anthocyanins.
