目的:采用代谢物组整体效应,考察不同剂量葛根芩连汤对链脲佐菌素(STZ)诱导糖尿病大鼠模型大鼠治疗作用的量效关系。方法:血浆预处理以后,UHPLC/Q-TOF采集血浆样品数据,建立数据矩阵,使用PCA等化学计量学的方法分析数据矩阵,提取能够反映病症与疗效的代谢物组,考察代谢物组随剂量的变化趋势,以代谢物组的葛根芩连汤整方的量效关系。结果:在主成分得分图中,剂量组随着剂量的增大,其样本点逐渐远离模型组向空白组回归;以剂量组主成分得分值拟合量效曲线图,得到S型曲线;量效参数为:量效关系剂量范围[D]0.2-[D]0.8=9.41-11.8g/kg,中位剂量:[D]0.5=10.55 g/kg,阈剂量:[D]0.2=9.41g/kg。结论:代谢物组可作为中药复方整体效应评价指标,为研究中药复方量效关系提供一种新的思路。
To investigate the dose-effect relationship of therapeutic effect of Gegenqinlian decoction(GGQLD) on STZ-induced diabetes on rats. Methods: After plasma pretreatment, the data of plasma samples was collected by UHPLC/Q-TOF to establish a data matrix, and analyzed by PCA, the metabonomics data was extracted to reflect the curative effect of GGQLD and the disease, inspected the change trend of metabonomic according to the change of dosage- and the metabonomies overall effect was used to evaluate the dose-effect relationship of GGQLD. Results: In PCA scores plot, as the dosage increased, the samples of administrated group moved away from the model samples, and returned to the control samples, and the principal component score values and the relevant dosages were used to fit the curve graph of dose-effect relationship, and the Sigrnoid curve obtained. The related parameters as follow: Effective dose range [ D ] 0.2 - [ D ] 0.8 = 9.41 -- 11.8g/kg, the median dose:[ D]0.5 = 10.55g/kg, threshold dose:[ D]0.2 = 9.41g/kg. Conclusion: The metabanomics could be used as an evaluation index for the overall effect of traditional Chinese medicine and provides a novel idea to study the dose-effect relationship of TCM.
