目的:观察氯化两面针碱、白花丹素、紫杉醇在体外对肝细胞的毒性作用。方法:大鼠肝脏经胰蛋白酶和胶原酶消化,进行体外肝细胞培养。经糖原染色法(PAS)鉴定后,采用MTT法检测不同浓度氯化两面针碱、白花丹素、紫杉醇对肝细胞的毒性作用。结果:氯化两面针碱、白花丹素、紫杉醇对肝细胞IC50分别为(3.501 5±0.672),(13.289 1±1.165)和(2.163 9±0.748)mg/L,且最高抑制率均超过80%,从而说明氯化两面针碱、白花丹素和紫杉醇均对正常肝细胞有毒性作用。结论:氯化两面针碱、白花丹素、紫杉醇可抑制肝细胞的增殖,对肝细胞有一定的毒性。
Objective:To detect the toxicity of nitidine chloride,plumbagin and taxol in rat's hepatocytes in vitro.Methods:SD rats' hepatocytes were cultured after digested with 0.25% trypsin,collagenase and identified by PAS.Hepatocytes proliferation was assayed by MTT method.Results:Treated with nitidine chloride,plumbagin and taxol,the 50 % inhibition concent-ration(IC50)of the hepatocytes was(3.501 5±0.672),(13.289 1±1.165),(2.163 9±0.748)mg/L;respectively and the biggest inhibitory rate over 80%,so nitidine chloride,plumbagin and taxol are cytotoxic in hepatocytes.Conclusion:Nitidine chloride,plumbagin and taxol have the function of inhibiting proliferation on the rats' hepatocytes.
