为探讨高脂日粮与基因型互作对血脂性状的影响,以高脂乳剂喂养200只小鼠1个月后,采用PCR-RFLP方法检测小鼠脂肪酸合成酶(FAS)基因型对其血脂水平和部分器官性状的影响。结果表明,高脂乳剂喂养1个月后,小鼠都患上了高脂血症。在对所有小鼠基因组DNA进行PCR扩增和限制性片段长度多态性分析,发现在所扩增的550 bp目的片段的27 486 082处(GenBank accession ref NT_039687.7)存在一个C/T的突变。相关性分析表明,TT基因型小鼠的TC、TG、FFA、心脏重、心/体重、脂肪重、脂肪重/体重明显高于CC基因型小鼠,差异显著(P〈0.05或P〈0.01);而TT基因型小鼠的HDL低于CC和CT基因型(P〈0.05)。结果提示,不同FAS基因型小鼠对高脂乳剂的敏感性存在一定的差异。
To investigate the effect of diet and gene on blood-fat traits of the individuals of the mOdel animals,200 mice were fed with high fat emulsion for one month ,then the genotypes of fatty acid synthesase were analyzed by PCR-RFLP to investigate their effect on blood-fat traits and some organ performances of mice. The results showed that, after fed with high fat emulsion for one month, all the mice suffered from hyperlipidemia. And a C/T mutation was found in 550 bp targeted fragments amplified at 27 486 082 site ( GenBank aeeession ref NT_039687.7), and association analysis showed that the mice carrying TT genotype had higher TC, TG, FFA, heart weight, heart weight/body weight, adipose weight, adipose weight/body weight than those of the mice with CC genotype ( P 〈 0.05 or P 〈 0.05 ), and the mice carrying TT genotype had lower HDL level than those of the mice with CC and CT genotypes ( P 〈 0.05 ). The results indicated that different FAS genotypes show different sensitativity to high fat emulsion in mice.
