目的通过对不同退变程度腰椎间盘的髓核细胞进行培养及活性测定,明确髓核细胞活性与MRI退变分型的相关性。方法取腰椎间盘髓核组织20例,均为我科收治单一手术节段患者,其中A组2例.为正常椎间盘髓核组织(PeaceⅠ&Ⅱ型);B、C、D三组分别6例,分别为MRI分型轻、中、重度(PearceⅢ型,Ⅳ型,V型),退变椎间盘髓核组织均在同样条件下进行体外髓核细胞的分离与原代培养,于5d,10d,15d,20d,25d五个时间段消化细胞,细胞计数了解各组细胞活性,CCK-8比色法绘制各组细胞的生长曲线,比较各组差异。结果髓核细胞计数在同时间点上A组明显高于B、C、D组,B、C、D组之间均存在显著统计学差异(P<0.01)。在同时间点上A组OD值(吸光值)明显高于B、C、D组,B、C、D各组之间OD值(吸光值)均存在统计学差异(P<0.01)。结论①正常腰椎间盘组织髓核细胞活性明显高于退变椎间盘髓核细胞。②MRI分型轻、中、重度退变腰椎间盘髓核细胞活性与其分型基本相符。
Objective To investigate the correlation between degeneration nucleus pulposus cell activity and MR imaging relative signal intensity and to identify the relationship of cells' activity and MRI features. Methods Twenty nucleus pulposus tissues from patients of our department and their surgeries all referred to only one intervertebral space. Group A were two normal nucleus pulposus tissues. There were 6 degeneration tissues in each group of B, C, and D, grouped by imaging type. In order to diminish the effect of age, we limited it from 35 yrs to 45 yrs. Nucleus pulposus cells were isolated from these tissues and cultured under the same conditions. We compared the generation one passage cells activity between different types. Results At the same time, between different imaging types, the cell counting had significantly statistical difference between group B and C, D (P〈0.01), and the cell counting of group A was obviously higher than that of group B, C, or D. The CCK-8 value of degeneration cells was significantly statistically different between group B and C, D (P〈0.01). Conclusions Normal lumbar disc nucleus pulposus cells' activity is more vigorous than degeneration cells'. MRI types of lumbar degeneration can basically reflect the activity of nucleus pulposus cells.
