目的研究枇杷叶三萜酸(TAL)对急、慢性炎症的抑制作用。方法采用二甲苯诱导的小鼠耳肿胀、冰醋酸诱导的小鼠腹腔毛细血管通透性实验、角叉菜胶诱导的大鼠足爪肿胀、棉球诱导的大鼠肉芽肿和福氏完全佐剂诱导的大鼠佐剂性关节炎模型,观察TAL的抗炎作用。结果TAL(75、225、675mg/kg)ig给药对二甲苯诱导的小鼠耳肿胀和冰醋酸引起的腹腔毛细血管渗出有明显的抑制作用;TAL(50、150、450mg/kg)ig给药可明显减轻角叉菜胶诱导的大鼠足爪肿胀、棉球诱导的大鼠肉芽肿和AA大鼠的原发性、继发性炎症病变。进一步研究表明,TAL(0.05、0.5、5、50mg/L)体外给药可增加ConA、LPS诱导的AA大鼠脾淋巴细胞增殖反应和IL-2的产生,同时抑制AA大鼠PM①产生过高的IL-1。结论TAL具有良好的抗炎作用。
Objective To study the anti-inflammatory effect of triterpene acids of loquat leaf (TAL). Methods To explore the anti-inflammatory actions of TAL, acute inflammatory models including mice ear edema, mice celiac capillary permeation, and rat palm swelling induced by dimethylbenzene, acetic, and carrageenin respectively and chronic inflammatory models including granulation hyperplasia and adjuvant arthritis (AA)caused by the implantation of cotton pill and freund's complete adjuvant( FCA) in rats respectively were used. Results TAL (75, 225, 675 mg/kg, ig) could significantly inhibit the ear edema and celiac capillary permeation in mice ; TAL (50, 150, 450 mg/kg , ig) could not only inhibit the paw swelling, granulation hyperplasia in rats, but also attenuate the primary and secondary inflammatory reaction, reduce the multiple arthritis score, and inhibit the hind-paw pathological changes in AA rats. TAL(0. 05, 0. 5, 5, 50 mg/L) could not only increase splenocytes proliferation and IL-2 production of splenocytes induced by ConA and LPS, but also reduce the elevated IL-1 production from peritoneal macrophage (PMΦ) in AA rats in vitro. Conclusion TAL has a remarkable anti-inflammatory effect.
