马氏正钳蝎β-肌动蛋白基因的克隆与系统进化分析

蝎子生物学研究主要包括蝎的解剖学,分类学,生物地理学以及与医学相关的毒液生物化学研究。在过去已构建的马氏正钳蝎(又称东亚钳蝎)毒腺c DNA文库的基础上,本研究克隆与鉴定了马氏正钳蝎的全长β-肌动蛋白基因的前体核苷酸序列。马氏正钳蝎全长β-肌动蛋白c DNA由1383个核苷酸组成,包括90个核苷酸5'非翻译区(5'UTR),147个核苷酸3'非翻译区(3'UTR)和1131个核苷酸的开放阅读框(ORF)。该马氏正钳蝎全长β-肌动蛋白前体核苷酸序列是首个从蝎目动物中分离鉴定的β-肌动蛋白c DNA序列。β-肌动蛋白基因系统进化分析显示蝎不仅是蛛形纲的原始类群,而且很可能是节肢动物的原始物种。该研究结果为蝎目的系统进化地位、甲壳-昆虫近缘假说提供了新的分子证据。

英文摘要:

For most of last century,the study of scorpion biology has been a phenomenological enterprise aimed primarily at descriptive anatomy,taxonomy,and biogeography,accompanied by detached medical research on venom biochemistry. In this investigation,based on the venomous c DNA library of the scorpion Mesobuthus martensii Karsch( synonym: Buthus martensii Karsch,Bm K),a new β- actin gene was cloned and characterized from Bm K. The full- length β- actin gene is comprised of 1383 nt,including 90 nt 5'UTR,147 nt 3'UTR and 1131 nt ORF. The β- actin gene from the species Bm K was firstly isolated and characterized from the order Scorpiones. Phylogenetic analysis of β- actin genes clarified that the species Bm K was the original group of the class of Arachnida. Phylogenetic analysis of β- actin genes furtherly indicated that scorpion was possibly the primitive species out of the phylum of Arthropoda. The results provided new molecular evidence for the phylogenetic and systematical rank of the species scorpion,and for the hypothesis of a crustacean- hexapod affinity.
