为了研究石墨烯对Hep G2细胞毒性效应的时间依赖性特征,采用超高效液相串联飞行时间质谱仪(UPLC-Q-TOF/MS)对石墨烯干预的Hep G2细胞的代谢产物进行了代谢组学研究。通过代谢组学的代谢轮廓分析发现,在浓度为25μg·m L-1石墨烯干预Hep G2细胞时,多变量偏最小二乘法的得分图上呈现了明显的时间代谢差异,同时依据载荷图、VIP图、箱图等多变量和单变量分析方法,获得了影响细胞代谢差异的潜在生物标志物。依据质核比、保留时间、母离子、碎片离子以及查询的相关代谢产物数据库,解析出了9个化合物的结构。通过结合相关代谢数据库(KEGG、HMDB、Met PA等)的分析和研究,发现了三个被石墨烯扰动较大的细胞代谢通路。
In order to study the time-dependent cytotoxicity of Hep G2 treated with graphene,the UPLCQ-TOF / MS technique was employed to analyze the endogenous metabolites of Hep G2 cell line. By the virtue of score plot of partial least squares discriminant analysis( PLS-DA),it was found that metabolic peculiarity of Hep G2 cell treated with graphene at a concentration of 25 μg ·m L- 1was discrepancy in different time phases. Combined multivariate with univariate analysis,including loading plot,VIP score plot,as well as Box figure,potential metabolic biomarkers were acquired. The structures of nine potential biomarkers were identified according to m / z,retention time,precursor ions,fragment ions and the metabolic data base. By analyses of the data bases such as KEGG,HMDB,Lipidomics and Met PA,the disturbed metabolic pathways were found.
