采用均匀设计法,对影响PCR的主要因素,即退火温度、引物、Taq酶、二甲基亚砜(DMSO)、模板DNA进行了优化,得到人类SMN基因的最优PCR扩增体系。结果表明,PCR扩增条件和反应体系的最优组合为:退火温度61.6℃、Taq酶0.26μL、引物(10μmol/L)3.6μL、DMSO(100%)2μL、DNA 3.8μL。
Based on uniform design method, the critical factors implicating with PCR, such as annealing temperature,primer,Taq polymerase,DMSO,DNA,were optimized respectively and PCR amplifying condition of human SMN gene was established. The results shouwed that annealing temperature 61.6 ℃,Taq polymerase 0.26 μL, primer(10 μmol / L)3.6 μL,DMSO(100%)2 μL,DNA3.8 μL were designated to the optimized combination between PCR amplifying condition and reaction system.
