玻璃化保存方法操作方便、保存效果良好,近年来已被广泛用于血管、肌腱等生物软组织的深低温保存.基于QLV理论,采用新的参数拟合方法得到QLV模型特征参数,通过比较玻璃化保存前后QLV模型中特征参数的差异,研究玻璃化保存对跟腱松弛行为的影响,以期建立和完善玻璃化保存跟腱的粘弹性力学评价指标.本研究对玻璃化保存前后的肌腱做应变水平为2%的松弛实验,并成功地应用QLV模型分别描述了跟腱标准松弛实验的线性加载段和应力松弛段应力随时间的变化.结果表明:①经过1 200s的松弛,玻璃化保存前后跟腱的最终应力松弛水平相差不大(约4%);②针对QLV模型,充分考虑标准松弛实验中线性加载段的影响,建立了一种松弛全过程数据拟合方法,用模型很好地描述了整个松弛过程应力随时间的变化;③通过对比玻璃化保存前后QLV模型中各特征参数的变化,发现跟腱经过玻璃化保存后其刚性变强、弹性响应能力增加,而其粘性响应有差异;④用Wilcoxon符号秩检验法对玻璃化保存前后跟腱的松弛行特征参数进行统计学分析,结果表明保存前后其特征参数没有显著性差异(p〉0.05),说明所设计的玻璃化保存方案能够很好地保留跟腱的粘弹性力学性能.
Vitrification,with its advantage of easier operation and better outcomes,is widely used to cryopreserve blood vessels,tendons and other soft tissues in recent years.Effects of vitrification cryopreservation on the viscoelastic properties of rabbit achilles tendons were investigated.Stress-relaxation behaviors of both fresh and vitrification cryopreserved rabbit achilles tendons were experimentally examined.These experimental data were used in combination with the quasi-linear viscoelastic(QLV) theory,to characterize the reduced relaxation function,G(t)(described by constants a,b,c,d,g and h) and the elastic response σe(ε)(described by constants A and B) of the tendons.In order to evaluate the viscoelastic properties of vitrification cryopreserved achilles tendon with high reliability,a new numerical fitting method was developed and used for QLV theory based analysis.The essential influences of vitrification cryopreservation method on the viscoelastic behaviors of the tendons were available by analyzing the difference of these parameters between the fresh and the cryopreserved tendons.The results of the research include: ①There is no significant difference of relaxation level between the fresh and the vitrification achilles tendon(about 4%).②A new numerical scheme was developed to optimize the QLV model,which fully considers the influence of linear load section of standard relaxation and describe the stress-time relationship successfully.③By comparing the characteristic parameters of QLV model,it was found that vitrification cryopreservation had a significant influence on the tendinous elastic properties.④By analyzing the differences of the parameters between the fresh and the cryopreserved tendons,with the Wilcoxon signed rank test method,we found the characteristic parameters were no significant differences(p0.05) before and after vitrification cryopreserved.
