目的 观察化痰通瘀解毒方(Huatan Tongyu Jiedu Decoction,HTJD)水提取物对胃癌细胞SGC-7901E-钙黏蛋白(Epithelial cadherin,E-Cad)、基质金属蛋白酶-2(matrix metallopro-teinase 2,MMP-2)表达及其侵袭转移能力的影响。方法 分别用含化痰通瘀解毒方水提取物5、10、20、30、40μg/mL的培养基干预人胃癌SGC-7901细胞24h后,采用划痕实验检测人胃癌细胞SGC-7901的侵袭迁移能力,采用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测SGC-7901细胞中和E-Cad、MMP-2的表达水平。结果 HTJD 10、20、30、40μg/mL浓度组与空白对照组相比,划痕愈合距离(△d)明显减小(P〈0.05或P〈0.01);HTJD 10、20、30、40μg/mL浓度组SGC-7901细胞上清液中E-Cad比空白对照组含量升高;细胞上清液中MMP-2的含量与空白对照组相比明显减少(P〈0.05或P〈0.01)。结论 化痰解毒方可抑制人胃癌细胞SGC-7901侵袭和迁移能力,其作用机制可能与下调MMP-2表达水平,上调E-Cad的表达水平有关。
Objective To observe the effects of the aqueous extracts of Huatan Tongyu Jiedu Decoction on expression of Ecadherin and MMP-2 in gastric cancer cell line SGC-7901. Methods Gastric cancer cell SGC-7901 was intervented for 24h by the extract of different concentrations of 5, 10, 20μg/mL, 30μg/mL and 40μg/mL, respectively. Wounding-healing assay test was adopted to detect gastric cancer cell SGC-7901's capacity of invasion and matastasis. Enzyme linked immunosorbent assay(ELISA) is operated to detect the protein expression of matrix metallopro-teinase 2 (MMP-2) and E-cadherin in cells. Results The difference of cells' healing width between the treatment concentration groups of 101.μg/mL, 20μg/mL, 30μg/mL and 40μg/mL and the blank group showed statitically significant(P〈0.05, P〈0.01 ); the amount of E-cadherin of the treatment concentration groups of 10μg/mL, 20μg/mL,30μg/mL and 40μg/mL was higher than that of the blank group(P〈0.05 ) ; the amount of MMP-2 in cell snpematant of the treatment concentration groups of 10μg/mL, 20μg/mL, 30μg/mL and 40μg/mL was obviously lower than that of the blank group (P〈 0.05). Conclusion HTJD repress the capacity of gastric cancer cell line SGC-7901 on invasion and matastasis. Its mechanism is likely related to down -reulating the expression level of MMP-2 and increasing the expression level of E-cadherin.
