目的 研究胃癌及癌旁正常组织细胞膜中蛋白质及脂质的傅立叶变换红外光谱(FTIR)的光谱变化及与相应组织、细胞层次光谱变化的差异。方法提取肿瘤组织和癌旁正常组织的细胞膜,将细胞膜进行溴化钾压片,应用Nicolet Magna750型红外光谱仪对细胞膜的溴化钾压片进行红外光谱检测,获得细胞膜的光谱图,选择其特征性光谱与胃癌组织、胃癌细胞株的相应光谱进行比对。结果胃癌组织细胞膜的红外光谱特征与相应正常组织细胞膜光谱特征存在异同,且胃癌组织细胞膜与胃癌组织、胃癌细胞株的光谱特征存在异同。主要表现在与脂类相关的2955、2920、2850、1740cm^-1谱带的变化。站论胃癌组织及癌旁正常组织细胞膜上生物大分子红外光谱发生变化及与相应组织、细胞层次红外光谱之间存在差异,其丰富了生物组织红外光谱研究的内容。
Objective To study the protein and lipid in the membrane of gastric cancer tissues and normal tissues with Fourier transform infrared (FTIR) and explore the special spectral features of the membrane of gastric cancer tissues compared with the membrane of normal tissues, cancer cells and cancer tissues. Methods We extracted membrane from cancer tissue and normal tissue samples, sheeted them With KBr, observed them by FTIR and compared the spectral data of the membrane of gastric cancer tissues, the membrane of normal tissues, cancer cells and cancer tissues. Results Both similarities and differences were found in the infrared spectral features between the membranes of gastric cancer tissues and normal tissues. The spectral features of the membrane of gastric cancer tissues had similarities and differences with those of gastric tissues and grastric cell strain, which were shown in the changes of the 2 957, 2 927, 2 853 and 1 740 cm^-1 bands related to lipids. Conclusion The results suggest that the spectral features of the membrane of gastric cancer tissues are significantly different from those of the membrane of normal tissues, cancer ceils and cancer tissues, which has enriched studies on biological tissues infrared spectroscopy.
