目的探讨黄姜水提取物(WED)对由高脂高胆固醇致家兔动脉粥样硬化的影响。方法采用高脂高胆固醇饲喂法建立家兔动脉粥样硬化模型,分别测定正常对照组、高脂模型组、WED低、中、高剂量组血清中TC、TG、LDL-C、HDL-C、MDA的含量及SOD的活性,采用大体染色计算脂肪斑块面积、组织切片观察。结果 WED均能明显降低血清中TC、TG、LDL-C、MDA的含量(P〈0.01),并能升高血清中HDL-C的含量(P〈0.01)和SOD的活性(P〈0.05),中、高剂量组家兔粥样斑块腹主动脉前段脂质面积百分比值与高脂模型组比较具有统计意义,分别为40.71%±9.13%(P〈0.05)、35.24%±3.09%(P〈0.01)、76.54%±19.53%;病理切片显示:各用药组家兔主动脉病变程度均小于高脂模型组,其中高剂量组的AS病变程度最小。结论 DWS具有抗高脂饮食诱导家兔动脉粥样硬化的作用,其作用机制可能与其调节血脂与抗脂质过氧化有关。
OBJECTIVE To observe the preventive effects of water extracts from Dioscorea zingiberensis C.H.Wright(WED) on atherosclerosis(AS) formation in rabbits.METHODS Rabbits was administered with high fat and cholesterol diet to make the AS model.The rabbits were divided into 5 groups randomly:control group,model group,WEDⅠ-Ⅲ group(low,middle,high dosage of WED).The contents of TC,TG,LDL-C,HDL-C,MDA and the SOD activity in serum were detected.Percentage of plaques covering aortic intima(PA)was calculated by gross stain.The changes of coronary arteries were observed with microscope and incidence of atherosclerosis of small coronary arteries was calculated.The following parameters were evaluated by biochemistry methods of malondialdehyde(MDA) and superoxide dismutase(SOD).RESULTS Compared with the model group,the rabbits of three WED groups decreased the contents of TC,TG,LDL-C,MDA significantly(P0.01),and the content of HDL-C in WED Ⅲ group was increased significantly(P0.01),also the same as the SOD.PA was 40.71%±9.13%(P0.05),35.24%±3.09%(P0.01),76.54%±19.53% in model,WED Ⅲ and WEDⅡgroups,respectively.Degrees of AS formation in aorta of all WED groups were less than that in model group.CONCLUSION DWS can prevent AS formation by high lipid and cholesterol feeding.The mechanism may be concerned with blood-fat adjustment and auti-liopid peroxidation.
