DNA extracted with the CTAB method from different O. sinensis strains and different parts of natural O. sinensis specimens was compared, and the qualities of DNA solutions were tested by PCR amplification. The results showed that the CTAB method can extract DNA successfully from O. sinensis strains and different parts of O. sinensis. The ascospore-wall breaking method to obtain DNA directly from ascospores was firstly reported, and then the difference between this method and CTAB method was discussed. DNA got from the two methods can be used for amplification very well.
