目的探讨胞内视黄醛结合蛋白(CRALBP)在豚鼠离焦诱导型近视眼中的表达。方法选取3~4周龄有色豚鼠10只,右眼戴-8.00D透镜,左眼不戴镜作为对照眼。戴镜前和戴镜后第11d分别测量眼轴长度和屈光度;通过免疫组织化学和蛋白质免疫印迹的方法检测CRALBP在豚鼠诱导近视眼和对照眼的视网膜、脉络膜和巩膜中的表达。结果戴镜后第11d,模型眼与戴镜前相比屈光度平均增加了一3.17D,与对照眼比较差异有统计学意义(P〈0.01);眼轴长度在两组眼之间差异无统计学意义(P〉0.05)。免疫组织化学结果显示,与对照眼相比,诱导近视眼的CRALBP在巩膜中的表达明显降低,在脉络膜和视网膜中的表达升高。Westernblot结果和免疫组织化学结果一致,CRALBP在诱导近视眼巩膜中的表达明显降低。结论CRALBP可能在豚鼠诱导近视眼的发生以及巩膜重塑中起一定作用。
Objective Researches showed that cellular retinaldehyde binding protein (CRALBP) involved in the visual cycle in mammal. However,whether CRALBP is related to the pathogenesis of experimental myopia is under clear. This study was to investigate the protein expression of CRALBP in lens-induced myopia in guinea pigs. Methods Lens-induced myopia model was created by wearing -8. O0 D concave spherical lenses in 10 right eyes of 3- or 4-week-old guinea pigs, and the fellow eyes were as control eyes. The refraction of guinea pigs was detected before wearing of lens and on the 11 th day after wearing of lens, and axial length of two groups was measured using A ultrasound. The protein expression of CRALBP was detected in retina, choroids and sclera by immunohistochemistry and Western blotting methods. Results After 10 days of lenses wearing, the average diopter of the right eyes of guinea pigs was - 3. 17 D, showing a significant difference in comparison with control eyes ( P 〉 0.01 ). However, there were no significant difference in axial length between these two groups ( P 〉 0.05 ). There was less brown particles of CRALBP in retina and sclera and more positve particles in chroiod in control eyes,and the positive particles in retina and chroid were significantly increased in model eyes. The Western blotting assay showed that the protein expression of CRALBP was significantly decreased in the posterior sclera and increased in retina and choroids in lens-induced myopic eyes of guinea pigs. Conclusion CRALBP may play a role in the lens induced myopia and scleral remodeling.
