目的 探讨清脑通络汤对脑梗死小鼠脑组织中TGF-β/Smad通路的影响。方法 将32只雄性C57BL/6小鼠随机等分为大脑中动脉栓塞(MCAO)组、清脑通络汤组、假手术组和空白组,每组8只。MCAO组和清脑通络汤组均应用线栓法构建右侧大脑中动脉栓塞模型,随后MCAO组给予生理盐水1 mL/100 g灌胃,清脑通络汤组给予清脑通络汤1 mL/100 g灌胃,均3次/d;假手术组仅暴露颈内动脉后缝合皮肤,随后给予生理盐水1 mL/100 g灌胃,3次/d;空白组不进行任何操作。术后72 h,对4组小鼠进行神经功能缺损评分(mNSS),RT-QPCR法检测缺血半暗带脑组织(假手术组和空白组取相应位置脑组织)中TGF-β、Smad2、Smad4和p21表达情况。结果 MCAO组和清脑通络汤组mNSS评分均明显高于假手术组和对照组(P均〈0.05),清脑通络汤组mNSS评分明显低于MCAO组(P〈0.05)。MCAO组TGF-β、Smad2、Smad4表达水平均显著高于其他3组(P均〈0.05),p21表达水平均显著低于其他3组(P均〈0.05);清脑通络汤组除Smad2表达水平明显高于假手术组和空白组外(P均〈0.05),TGF-β、Smad4和p21表达水平与假手术组和空白组比较差异均无统计学意义(P均〉0.05)。结论 清脑通络汤可调控TGF-β/Smad信号通路相关基因的转录与表达,并通过提高靶基因p21的表达而对脑梗死小鼠起到一定的脑保护作用。
Objective It is to explore the impact of Qingnao Tongluo decoction on TGF-β/Smad signal pathway in the brain tissue of cerebral infarction mice model. Methods 32 male C57BL/6 mice were randomly divided into four groups equally,including middle cerebral artery occlusion (MCAO)group, Qingnao Tongluo decoction group, shame group and control group. Embolism models of right middle cerebral artery were established in MCAO group and Qingnao Tongluo group which were subsequently treated with normal saline 1 mL/100 g and Qingnao Tongluo decoction by lavage respectively three times per day. The skin was closed after internal carotid artery was exposed in sham operation group which was subsequently treated with normal saline 1 mL/100 g by lavage. The control group was conducted with no operation. At 72 h after operation, neurological function was evaluated according to modified Neurological Severity Score (mNSS) , the expression level of TGF-β, Smad2, Smad4 and p21 in ischemic penumbra of cerebral tissue (respective position of cerebral tissue in sham operation group and control group) was measured by RT-QPCR. Results The mNSS of Qingnao Tongluo decoction group and MCAO group was significantly higher than that in sham operation group and control group (P 〈 0.05), furthermore, mNSS in Qingnao Tongluo decoction was lower than that in MCAO group(P 〈 0.05). The expression of TGF-β, Smad2, Smad4 and p21 were significantly higher while that of p21 was lower in MCAO group than that in the other three groups( P 〈 0.05) ; the expression of Smad2 was significantly higher in Qingnao Tongluo decoction group than that in sham operation group and control group(P 〈 0.05 ), but there was no significant difference in the expression of TGF-β, Smad4 and p21 among Qingnao Tongluo decoction group, shame operation group and control group (P 〉 0.05).
