目的:建立体外培养sD大鼠小脑神经元划痕模型。方法:用200μl塑料枪头划痕体外培养大鼠小脑神经元,观察相同视野下不同时间点(0,24,48h)的划痕宽度。结果:倒置相差显微镜下可见划痕细胞后0h即形成划痕区,边缘整齐。24h后,划痕边缘变模糊同时划痕宽度变窄。48h后,划痕宽度进一步缩小。结论:本模型简便经济,能较好地模拟脑损伤后神经元的迁移。
Objective : To establish a scratch assay model by using the cerebellar neurons of SD rats in vitro. Methods: Scratch was induced by a 200μl plastic pipette tip on cerebellar neurons of rats in vitro. The widths of scratch at different time points (0 h, 24 h, 48 h) from the same field were measured. Results: An artificial scratch was created and the edge of the scratch was smooth at 0 h after scratching cell under an inverted phase microscope. At 24 h, the edge of the scratch blurred and the width became narrower. At 48 h, further reduction of the width of the scratch was observed. Con- clusion: This model proved to be a convenient and cost-effective method that can well mimic the cerebellar neurons migra- tion after brain injury.
