目的研究五味子醇提取物减轻丙戊酸钠所致肝损伤的作用及协同抗癫痫作用。方法第一部分实验动物随机分为:空白对照组,模型组,五味子醇提取物低剂量组,五味子醇提取物高剂量组。连续给药14 d后检测动物血清中丙氨酸氨基转移酶(ALT),天冬氨酸氨基转移酶(AST)含量。酶联免疫吸附测定(ELISA)检测肝组织中谷胱甘肽过氧化物酶(GSH-Px)和超氧化歧化酶(SOD)等含量。第二部分实验动物随机分为:模型对照组,五味子醇提取物组,丙戊酸钠组,丙戊酸钠联合五味子醇提取物组。实验动物连续给药7 d后,按照经典癫痫发作行为标准评价五味子醇提取物的协同抗癫痫作用。结果五味子醇提取物高剂量组血清ALT、AST含量分别为(45.60±10.54)、(154.60±33.46)U/L,与模型组比较显著降低(P〈0.05)。五味子醇提取物高剂量组肝组织中的SOD,GSH含量分别为(4.50±1.16)U/mL、(47.53±5.11)g/L,与模型组比较显著升高(P〈0.05)。结论五味子醇提取物可能通过增加肝脏中的GSH及SOD含量来降低小鼠血清中ALT、AST。五味子醇提取物能够减轻小鼠癫痫发作症状。
Objective To study the Schisandra Ethanol Extract reduce liver injury induced by sodium valproate and the of effect synergistic antiepileptic.Methods The first parts of the experimental animals were randomly divided into: blank control group,model group,schizandrol extract low-dose group,schizandrol extract high-dose group.After 14 days continuous administration animal serum alanine amino transferase enzyme(ALT) and aspartate aminotransferase(AST) content was detected.ELISA detected liver tissue glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) content.The second parts of the experimental animals were randomly divided into: model control group,the schizandrol extract group,sodium valproate group,sodium valproate joint Schisandra extract group.Experimental ani mals administered continuously for 7 days,according to the classic epileptic seizures standards of behavior evaluation schizandrol extract synergies antiepileptic effect.Results Schisandra Ethanol Extract high-dose group serum ALT,AST content were(45.60±10.54) U/L and(154.60±33.46) U/L,the comparison with the model group was significantly lower(P 0.05).Extract schizandrol high dose group in the liver tissue SOD,GSH levels were(4.50 ±1.16) U/mL,(47.53±5.11) g/mL,the comparison of its value with the model group were significantly different(P 0.05).Conclusion The extract of Schisandra Ethanol may increases the GSH and SOD levels in the liver to reduce serum ALT and AST.The Schisandra Ethand Extract can alleviate mice symptoms of seizures.
