目的:研究能量可控陡脉冲(ECSP)对荷瘤大鼠淋巴细胞活化及细胞因子的影响,探讨ECSP治疗肿瘤的作用机制。方法:采用细胞悬液注射法建立Wistar大鼠皮下瘤模型,ECSP处理后每3天检测肿瘤大小。处理后2周,利用MTT法检测各组大鼠脾淋巴细胞增殖反应及淋巴细胞活性,ELISA法检测大鼠脾淋巴细胞分泌的IL4和IFN-γ的表达。结果:ECSP处理组肿瘤生长明显受到抑制;ECSP处理组淋巴细胞增殖反应、淋巴细胞活性的表达明显高于荷瘤对照组(P〈0.05,P〈0.05),荷瘤对照组的细胞因子IFN—γ呈低表达,而IL-4呈高表达,与正常对照组比较差异显著;ECSP处理后,与同期荷瘤大鼠对照组比较IFN-γ表达水平升高,而IL-4表达水平降低。结论:ECSP可以抑制肿瘤生长,诱发机体抗癌免疫反应,改善和恢复荷瘤鼠淋巴细胞的免疫活性。
Objective :To investigate the effects of immune function of energy controllable steep pulse on the transplanted tumor in rat. Methods:Thirty wistar rats were experimented. Tumor size was measured before and after treatment to evaluate the reaction of the treatment. After two weeks of treatment, MTT methods were used to observe the lymphocytes proliferation and activity,cytokines expression levels include IL-4 and IFN-γ were measured by ELISA. Results:Growth of the tumor was significantly inhibited in ECSP therapy group as compared with the controls. It was founded that the Lymphocytes proliferation, Lymphocytes activity in ECSP therapy group was similar to that of the normal control group, there was no obvious difference( P = 0. 953, P = 0. 130), but these parameters were markedly higher than those of the control group(P 〈0. 05 ,P 〈0. 05). IFN-γ level in rat with subcutaoneous transplantive tumor was lower than that of normal rats, but IL-4 level was higher than those of normal rats. After ECSP, expression of IFN-γ, level increased significantly(P 〈0. 05), however, expression of IL-4 level decreased obviously compared with those of control group. Conclusion: It shows that ECSP could inhibited the growth of the tumor and induce the host immune reaction of anti-tumor efficiently and improve the immune activity of lymphocyte in rats with tumor.
