从北京林业大学校园和海南省儋州植物园共采集6种植物的不同组织,通过五步法表面消毒和选择性分离培养方法共分离得到183株植物内生菌,平板拮抗试验显示,32株内生菌对大豆菌核病病原菌具有拮抗活性,占分离菌株的17.49%;结合抗菌活性、形态特征和宿主来源,选择10株拮抗菌株进行鉴定,结果显示这些菌株分别属于链霉菌属(Streptomyces)、棒状杆菌属(Microbacterium)、根瘤菌属(Rhizobium)、无色杆菌属似(Achromobacter),具有丰富的微生物多样性。因此,本研究筛选的大豆菌核病病原菌拮抗菌株为开发生物防治混合菌剂提供了菌种资源,为生物防治菌核病、减少农业经济损失奠定了基础。
Using selective media, a total of 183 endophytes were isolated from surface sterilization tissues of 6 plants collected from the campus of Beijing Foresry University and the botanic garden in Danzhou. Antagonism assays by dual culture were performed that 32 strains, 17.49 % of all isolates, presented good levels of inhibition against S. selerotiorum. Combined with the inhibit activity, morphological characterization and host, 10 endophytes were selected to further identify by 16S rRNA gene sequencing. The results permitted assignment these antagonistic endophytes to 5 different genera, Streptomyces, Microbacterium, Rhizobium, Achromobacter and Bacillus with a level of diversity. Therefore, the antagonistic endophytes screened in this study will provide new resources and lay the foundation to exploring mixture microorganisms for biocontrol of Sclerotinia selerotiorum and enhance of agricultural produetion.
