采用5.0L全自动发酵罐进行钝顶螺旋藻培养的研究

本文采用5.0L全自动机械搅拌式发酵罐作为培养装置,分别进行了钝顶螺旋藻光合自养和混合营养培养研究。通过测定最终藻体细胞中藻体干重(DW)、藻胆蛋白和叶绿素a等成分,可知混合营养条件下,藻体干重比光合自养培养时提高104.75%,但藻胆蛋白和叶绿素a的含量分别低13.5%和18.2%。在此基础上,进行了螺旋藻混合营养条件下半连续培养研究,结果表明:藻体干重比采用混合营养分批培养时提高38.05%,而藻胆蛋白和叶绿素a的含量则分别低3.76%和1.75%。从最终培养结果可知,在规模化培养时利用光生物反应器进行半连续培养是一种可行的培养方式。

In this paper, a 5.0L automatic fermenter was used respectively to cultivate the S. platensis in the condition of photoautotrophy and mixotrophy, Three critical ingredients were measured timely in the process of the cultivating S. platensis under photoautotrophy and mixotro-phy, including dry weight (DW), phycobiliproteins and chlorophyll-a. According to the experi-ment results, the DW that was in mixotrophy condition was 104.75 more percent than the pho-toautotrophy; the phycobiliproteins and the chlorophyll-a yet were lower, whose value were 13. 5 percent and 18.2 percent, respectively. Then the semi-continuous cultivation of S. platensis was researched. The results showed that the DW was 38.05 more percent than batch cultivation. However, the phycobiliproteins and the chlorophyll-a yet were lower, ; the value was 3.76 per-cent and 1.75 percent, respectively. The final cultivating results showed that the semi-continu-ous cultivation mode using photobioreactor was an effective method when cultivated in large-scale.
