目的分离鉴定中华隐孔菌(Cryptoporus sinensis Sheng H.Wu & M.Zang)子实体的化学成分。方法采用硅胶柱色谱、ODS柱色谱、Sephadex LH-20柱色谱、HPLC等分离方法,对中华隐孔菌子实体甲醇提取物的二氯甲烷萃取部分进行分离,根据理化性质和波谱数据鉴定化合物结构。采用MTT法对化合物进行细胞毒活性测定。结果分离鉴定了4个已知的隐孔菌酸化合物,分别是隐孔菌酸B(1)、隐孔菌酸B甲酯(2)、隐孔菌酸G(3)、1′,1″-二羧基隐孔菌酸D(4)。抗肿瘤活性筛选表明,化合物1~4具有弱的细胞毒活性,对肿瘤细胞PC-3、PANC-1、A549和MCF-7的半数抑制浓度大于100μmol.L-1。结论化合物1~4均是首次从该种真菌中分离得到。
Inedible mushrooms have been demonstrated to be a rich source of biologically active secondary metabolites.Cryptoporus sinensis as a new species inedible mushroom was separated from Cryptoporus volvatus in 2000 for the first time.Previous investigations focused on Cryptoporus volvatus,which some antitumor substances were isolated.This promoted us to investigate bioactive constituents from Cryptoporus sinensis.The MeOH extract of the fruiting bodies of Cryptoporus sinensis was dissolved in H2O and partitioned with CH2Cl2.The CH2Cl2 portion was subjected to silica gel,ODS,LH-20 chromatography and finally purified by reverse-phase HPLC.As a result,four known cryptoporic acid compounds,cryptoporic acid B(1),cryptoporic acid B methyl ester(2),cryptoporic acid G(3),1′,1″-dicarboxylic cryptoporic acid D(4),were obtained.Their structures were determined on the basis of physicochemical and spectral data.Using MTT assay for biological screening,compounds 1-4 showed weak cytotoxic activity to tumour cell PC-3,PANC-1,A549 and MCF-7(IC50100 μmol · L-1).
