目的探讨解整合素-金属蛋白酶33基因(ADAM33)F1、L1、T1位点单核苷酸多态性与新疆维吾尔族成人支气管哮喘的相关性。方法采用限制性片段长度多态性方法分析206例维吾尔族成人支气管哮喘患者(哮喘组)与140例维吾尔族成人健康体检者(对照组)ADAM33基因F1、L1、T1位点单核苷酸多态性,并对其中部分进行基因检测验证。结果两组ADAM33基因T1位点3种基因型分布差异有统计学意义(χ^2=10.472,P=0.005),等位基因频率分布差异有统计学意义(χ^2=11.192,P=0.001,OR=1.883,95%CI=1.296~2.737)。与AA基因型比较,G等位基因的携带者(AG、AG+GG)基因型均能明显增加哮喘发生的危险(P分别为0.003、0.009,OR分别为3.889、1.815,95%CI分别1.523~9.928、1.156~2.850)。结论 ADAM33基因T1位点单核苷酸多态性与新疆维吾尔族成人支气管哮喘具有明显相关性。
Objective To investigate the correlation of the F1,L1,T1 locus in ADAM33 gene polymorphism with bronchial asthma of Uyghur adults in Xinjiang.Methods PCR-RFLP was used to determine polymorphism of F1,L1,T1 locus in ADAM33 gene in 206 Uygur patients with bronchial asthma(asthma group)and 140 Uygur healthy individuals(control group),in which a part of the samples were detected for genetic verification.Results The comparison of three genotypes of the T1 in ADAM33had statistical significance in asthma group and control group(χ^2=10.472,P =0.005).Allelic frequencies had statistical significance.Compared with"AA",G allele carriers(both AG and AG+GG)could elevate the risk of asthma occurs(P =0.003,0.009;OR =3.889,1.815,95%CI =1.523-9.928,1.156-2.850).Conclusion The polymorphism of the T1 locus in ADAM33 gene possessed significant correlation with Uygur population in Xinjiang.
