目的:明确甘氨酸对缺血缺氧心肌细胞具有保护作用。方法:建立心肌细胞缺血缺氧模型,施加甘氨酸干预,应用高效液相色谱观察心肌细胞ATP、ADP、AMP含量变化及ATP/ADP比值。结果:模拟缺血缺氧1h后开始心肌细胞ATP含量较正常组明显降低(P〈0.01),而ADP、AMP含量与ATP变化相反,ATP/ADP比值较正常组也明显降低(P〈0.01);甘氨酸干预复合缺血缺氧后,心肌细胞ATP含量及ATP/ADP比值较单纯缺血缺氧组显著降低(P〈0.01),而ADP、AMP含量明显升高(P〈0.01)。结论:甘氨酸可以明显改善缺血缺氧心肌细胞的能量储备,发挥其保护效应。
Objective: To investigate the protective effects of glycine in cardiomyocyte subjected on ischemia and hypoxia. Methods: Cardiomyocytes model were established in vitro. The content ofATP, ADP and AMP were tested by high performance liquid chromatogram (HPLC). Results: The content of ATP of cardiomyocytes subjected to ischemia and hypoxia were significantly lower (P〈0.01) after 1 h. On the contrary, the eontent of ADP and AMP were significantly higher. The change of ATP/ADP was similar with ATP. However, when cardiomyocytes were subjected to glycine (5 mmol/L) combined with ischemia and hypoxia, the content of ATP and ATP/ADP were increased and the content of ADP and AMP were significantly decreased than those without treatment of glycine (P〈0.01). Conclusion: Glycine can improve obviously the energy metabolism of cardiomyocytes subjected to ischemia and bring into protection effect.
