整合素α2β1对肝癌细胞粘附与趋化行为的影响

采用微吸管技术,研究整合素α2β1对肝细胞癌(HCC)细胞SMMC-7721与Ⅳ型胶原衬表面粘附力特性和该细胞对Ⅳ型胶原趋化行为的影响.观察Anti-α2、Anti-β1对SMMC-7721细胞与Ⅳ型胶原裱衬表面的粘附力的影响;采用双微吸管实验法,观察SMMC-7721细胞伪足形成过程以及Anti-α2、Anti-β1对SMMC-7721细胞伪足形成的影响.利用流式细胞仪对SMMC-7721细胞表面整合素α2、β1亚单位的表达进行分析.结果表明,SMMC-7721细胞与5μg/mL Ⅳ型胶原裱衬表面之间的粘附力为952±134(×10^-10 N,n=60),加入5μg/mL Anti-α2粘附力减小到605±128(×10^-10 N,n=60);加入10μg/mL Anti-α2时粘附力减小到579±57(×10^-10 N,n=50).加入5μg/mL Anti-β1粘附力减小到449±119(×10^-10 N,n=60);加入10μg/mL Anti-β1时粘附力减小到220±78(×10^-10 N,n=55).双微吸管趋化实验表明:两侧微吸管加入相同浓度Ⅳ型胶原,细胞向两侧微吸管均有伪足形成;在此基础上,微吸管分别加入Anti-α2、Anti-β1的一侧,SMMC-7721细胞伪足生长曲线呈现明显的抑制,而未加入抗体的微吸管一侧与加入的对侧相比,该侧细胞的伪足明显增加.流式细胞仪分析表明,所研究细胞整合素α2、β1亚单位表达率达分别为23.17%和95.78%.上述结果表明,整合素α2β1是联系SMMC-7721肝癌细胞与Ⅳ型胶原裱衬表面粘附以及SMMC-7721细胞对Ⅳ型胶原趋化性伪足形成的重要受体基础.

英文摘要:

To quantitatively describe the effect of integrinα2β1 on the adhesion of hepatocellular carcinoma (HCC) cells (SMMC -7721) to type Ⅳ collagen (Col Ⅳ) coated surface and the chemotaxis of SMMC -7721 to Col Ⅳ. A micropipette technique was adopted to investigate the effect of blockade of integrinα2β1 on adhesion of SMMC -7721cells onto Col Ⅳ coated surfaces and pseudopod protrusion of SMMC -7721 cells in response to Col Ⅳ stimulation. Adhesion strength was expressed. Chemotactic pseudopod protrusion of a SMMC -7721 cell was evaluated using a dual - pipette set - up, in which two pipettes filled with Col Ⅳ solution were positional in close contact with the same cell. The lengths of pseudopods were measured and plotted against time to obtain a pseudopod growth curve. The integrinα2β1 on the surfaces of SMMC -7721 cells was analyzed with flow cytometry. The results show that the adhesion forces for SMMC - 7721 cells adhering on 5 μg/mL Col Ⅳ coated surfaces were 952±134(×10^-10 N, n =60). Upon treatment of SMMC - 7721 cells with Anti - α2 in a protein concentration of 5 μg/ml and 10μg/mL , the value decreased to 605±128 (×10^-10 N, n = 60) and 579±57 (×10 ^-10 N, n = 50) respectively. In addition, upon treatment of SMMC - 7721 cells with Anti - β1 in concentration of 5 μg/ml and 10 μg/ml, the value decreased significantly to449±119( × 10^-10 N, n = 60) and 220±78( ×10^-10 N, n =55), respectively. In dual pipette chemotaxis experiment, when the two pipettes were filled with Col Ⅳ, pseudopods extended from the SMMC -7721 cell into each of the pipettes nearly symmetrically, i. e. , with nearly identical maximum pseudopod length and similar pseudopod growth curves. Upon addition of Anti - α2 to one of the pipettes, pseudopod protrusion was blocked while protrusion into the opposite pipette became more evidently. In the same way, upon addition of Anti - β1 to one of the pipettes, pseudopod protrusion was also blocked nearly completely while protrusion
