由于方法的不同,人类多能干细胞(h PSCs)在体外产生的各种造血细胞的成熟度和功能存在偏差。我们实验室建立了高效的h PSCs与造血微环境基质细胞共培养产生大量造血干/祖细胞的方法。本专题就2种自然免疫相关细胞的早期发育和成熟,以及初期原始造血和成体造血的比较,建立了时序性地观察了h PSCs向造血细胞逐级诱导分化的方法 。
Among different culture systems, there exists clear discrepancies in maturity and function of hematopoietic cells induced from human pluripotent stem ceils (hPSCs). We have established an efficient system to produce large-scale hematopoietic stem/pregenitur cells by co-culturing hPSCs with stromal cells from definitive hemotopoietic niches. In this special topics, we present our recent data on the establishment of the method to induce hPSC-derived innate immune cells with functional maturity (eosinophils and macrophages), We also show our study on human very eraly hematopoiesis, representing the primitive wave, by embryoid body method, as well as on those derived from definitive ones. Our serial observation on hPSC-derivod hematopoietic cells should help in further recognition of the mechanisms controlling human early hematopoiesis and may benefits the future clinical application.
